This is a HeLa cell-based reporter kit designed to allow the customer to screen or identify agents (drugs, natural products, small molecules, synthetics, miRNAs, and genes) that affect or impact the process of NHEJ DNA repair. The kit uses GFP as an in vivo readout for the NHEJ pathway and a plasmid expression vector containing the mega-endonuclease I-Sce1.  The GFP gene is constructed in such a way that following transfection by I-Sce1 mega-endonuclease, any cells under NHEJ (either classic or alt) will appear as GFP+ cells.  The %GFP readout is a measure of NHEJ efficiency

1. HN-HeLa Cells ready for subculture.
2. I-Sce1 gene expression plasmid, pSCE (store at -20^o or 4^oC). See tube label for DNA concentration. We supply 50ug of pSCE for the base 25 assay kit. Larger kits are also available for labs requiring more I-Sce1 expression plasmid.  Note that we steeply discount plasmid DNA when purchased as a kit component.
3. Neomycin (50mg/ml stock, 1ml).  For routine maintenance of cell lines, use Neo at 200ug/ml (4ul per ml dilutions into medium+ serum).
