This is a HeLa cell-based reporter kit designed to allow the customer to screen or identify agents (drugs, natural products, small molecules, synthetics, miRNAs, pathway, signaling or response genes) that affect or impact the process of Homology Repair (HR). The kit uses GFP as an in vivo readout for the HR pathway. The HeLa cells included in this Kit contain stably integrated GFP plasmid which is mutated and therefore GFP minus. Following HR repair, the GFP gene converts to wild type GFP and the cells fluoresce. HR is initiated by transfection with an I-Sce1 Expression plasmid (included). Analysis of pooled transfected cells reveals GFP+ cells in 2-3d post-transfection. GFP+ cells have successfully completed HR and can be tracked with single cell resolution.

This means that the results will be highly physiological because events are transpiring within the cell in real time. Kit Contents
- HR-HeLa Cells frozen (cyro-protective medium) with the endogenous HR GFP reporter stably integrated. Cells may be sent frozen or ambient.
- I-Sce1 gene expression plasmid (50ug).
- Puromycin Stock.
- A detailed protocol describing how to set up and use this kit for assaying HR mechanisms in the cell host provided.
- Customer should specify the Kit size (25, 50, 100 assays corresponding to 50, 100 or 250 ug of I-Sce1 plasmid DNA, respectively).
- Optional: add WT GFP (e-GFP) as a control plasmid to this kit (50ug).
